Yes, it includes the base game and the DLC on the cartridge as opposed to the previous release that’s just the base game

No, sadly, it does not. It just has the cart and some interior artwork.

Q: i don't see the DLC content when i boot up, do i have to clear the main story first?

You have to play the game to unlock characters. The DLC is builded into the game.
