Will this fit over an otter box?

It is very stretchy. I don't think you will have a problem. I have a Galaxy S8 with a case, and although not an Otter Box, is still larger than the phone w/o a case and there is no problem

is there a locking method to prevent rotation? (specs state 360 degree rotation)

It doesn't move unless you move it yourself. I ride through bumps and so far its been steady I haven't had one issue.

how wide is the bar connection?

There is an adjustable strap that wraps around most any size handlebar and it is about 1/2" wide. Hope this is what you were wanting to know.
