Does this tape have a clear overlay I'm assuming that it does

Mine had an adhesive backing that peals off to install. Then a clear overlay that you peel off when you get the tape installed. Be careful when you peel both. Take your time. It will be worth it.

I have a 2016 Nissan Frontier pick up. Is this long enough to go from front to back?

I have trouble separating the clear film when I need to remove it. Any tips to make it easier?

Start at the cut end. Made it longer than you need. Cut the doors and the ends after you pull it off. Watch the video at the url I have on the site and the cards I sent with the stripes.
