The bristles are approximately 2 1/2 inches long, and medium stiffness.

Are the bristles natural or synthetic?

The label says “dense and durable PET bristles which are gentle and won’t scratch the cleaning surface.” I was astounded at how effective this brush is in getting everything up with one sweep. I had the label handy because I got another one for a friend who always has crumbs on her table which she tries to get up by hand.

Does the black loop for hanging bother your hand while using this, or feel uncomfortable?

The only time i notice the loop is when hanging up the broom or while spinning it around my finger. Its just a shoelace-like string so it's never an inconvenience
