Q: Can this 6" damper connect directly to the ac infinity cloudline t6 fan or it need to connect to duct first then fan?

No. This has a female sized connection. The fan's diameter is for attachment of ducting. It is the SAME SIZE as the damper. A short section of duct will be needed to connect the two. Could use a 3 inch piece of 6 inch rigid duct and tape it.

Q: Will this work on the intake side of an ac infinity fan? i want to use this to pull air from a room and not allow the air to flow back into that room

It will work on both the intake and exhaust side of the fan. I use it as part of a room exhaust system in my setup, it blocks the backdraft of air but if will let some heat or cold radiate through the metal flaps.
