How difficult is it to replace the seals?

280 no problem if you know your way around an engine, 350 is quite a bit more difficult due to the space you have to work with..

Does the cylinder have to be removed from the car to replace the o-ring?

You can do it by removing the hydraulic hoses as far back toward the pump giving enough slack to get the cylinder through the window and work on it
