Does this work for multiple lines? Are you able to put someone on hold without just muting them?

This does not put lines on hold, you would still have to do that from your phone set. This device picks up the handset mechanically to answer.

Will this work with a Jabra Engage 75 Stereo headset?

I plugged one of these into an Engage 65 base station, which should work the same, and it worked fine. Just adjust the knob so the handset doesn't fall off, make sure the ring detector is properly over the speaker (the round slider deal), and then if the red light is flashing, press the button at the bottom to enable ring detection.

I have Mitel 5330e phone and Jabra 9450 will this lifter work OK with this phone and headset?

Yes. The lifter will come off the back of the 9450 base and attach to the phone.
