Will these spindles fits a 4x4?

No. The spindle is for 2x4 only. A 4x4 spindle would have a hole in the center for a cv axel to slide through.

Does this kit work on a 2001 2WD Expedition

The spindles will fit the front, but I forget if the rear is leaf spring or coil spring.

How big are the blocks?

I had to use actual 2inch blocks to make it level. They’re closer to like 1.5 inches. It squatted a bit with the blocks in the kit. Luckily a friend had taller blocks.
